Cartographie golgienne des enzymes de synthèse des polysaccharides complexes pariétaux et recherche de complexes enzymatiques.

Abstract : Primary cell wall of dicotyledonous plants is composed of cellulose microfibrils, proteins, and complex polysaccharides represented by hemicelluloses and pectins. In Arabidopsis thaliana plant model, xyloglucan (XyG) and rhamnogalacturonan-I (RG-I) represent the main hemicelluloses and pectins polysaccharides respectively. Before cell wall transport, XyG and RG-I are synthesized in the Golgi apparatus, particularly via sugar interconversion enzymes and glycosyltransferases, which will generate and assemble the different sugars to form polysaccharides.The aim of this research project was to localize in Golgi subcompartments different cell wall complex polysaccharides epitopes and enzymes implicated in XyG and RG-I synthesis, as well as enzymatic complexes research, using confocal laser scanning microscopy, transmission electron microscopy and quantitative immunocytochemistry.
