Biological indicators of occupational exposures and related effects in farmers

Résumé

The aim of this project is to identify biomarkers of occupational exposures and the related effects in agricultural workers by using biobanks. This project will be divided into three parts: quantification of biological exposure indicators, analysis of DNA damages and global metabolic modifications associated.The first part of the study will be to select relevant pesticides according to their uses, their presence on the market, their toxicokinetic data and their natural degradation. The selected compounds, their metabolites and their degradation products will then be quantified in urine and plasma samples by LC-MS-MS and GC-MS-MS in biobanks samples. Secondly, the DNA obtained from the leucocytes of agricultural workers will be analysed by LC-MS-MS. An analysis method will be set up to allow the detection of 17 compounds. This method will highlight epigenetic and genotoxic alteration and will be correlated to exposure data. Finally, a metabolic profile as exhaustive as possible will be acquired for each individual from the biobanks from urine and plasma. Relative concentrations of these compounds will then be compared between the workers according to potential exposition using statistical analysis.Thanks to these analyses, the impregnation level of agricultural workers will first be determined. Then, the impregnation levels will be associated with DNA damages profiles, giving evidences of the effect of agricultural tasks of concern. Then they will be included in metabolomic analysis to determine new effect biomarkers.
